Assay Rapidly Responds to Viability Changes The results in Figure 3 illustrate the immediate response of the luminescent signal to changes in cell viability. In this example iCell® Cardiomyocytes were grown in medium containing the RealTimeGlo™ reagents. After 2 days, digitonin was added. The data show that as soon as the cells die, the luminescent signal decreases. 1,000,000 Add digitonin 100,000 10,000 1,000 01020304050 Time (hours) Figure 2. Effect of digitonin on iCell® Cardiomyocytes. Upon addition of digitonin, cell death is indicated by an immediate decrease in luminescent signal. The Main Points The ability to measure viability continuously in the same well over a 3-day period provides kinetic information on viable cell number during the course of an experiment and enables real-time decision making in reaction to observed viability changes. The simple assay protocol saves time and labor, and the ability to monitor viability continuously without sacrificing cells creates efficiencies by saving plates and reagents and reduces errors from preparing replicate samples.
