Luminescence (RLU) 13132MA The Caspase-Glo® 1 Inflammasome Assay provides a luminogenic caspase-1 substrate, Z-WEHD-aminoluciferin, in a lytic reagent optimized for caspase-1 activity and luciferase activity. A single addition of the reagent results in cell lysis, substrate cleavage by caspase-1 and generation of light by Ultra-Glo™ Recombinant Luciferase. Inclusion of the proteasome inhibitor MG-132 in the reagent eliminates nonspecific proteasome-mediated cleavage of the substrate, enabling sensitive measurement of caspase-1 activity. The caspase-1 selective inhibitor, Ac-YVAD-CHO, can be added in parallel wells to confirm caspase-1 specific activity. Benefits: • Quick, Easy-to-Perform Assay: No sample preparation required. Add the Caspase-Glo® 1 Reagent directly to cells or culture medium in multiwell plates, and measure luminescence after only 1 hour. Less time and labor required than for Western blot or ELISA. No need for multiple pipetting steps. • Accurate Determination of Caspase-1 Activity: The assay measures only catalytically active caspase-1, enabling precise time courses of enzyme function. Westerns and ELISAs don’t necessarily monitor active enzyme. Citation Available: Read a publication by an early tester.
